Description:
Bovine β2-glycoprotein 1 (β2-GP 1), also known as Apolipoprotein H or Apo H. Natural antigen for solid (ELISA) and fluid phase diagnostic assays.
Immunological function:
Binds IgG, IgA and IgM-type human auto-antibodies. Auto-antibodies to β2-glycoprotein 1 recognize conformation-dependent and linear epitopes.
Origin:
Native. Purified from fetal calf serum (FCS) by protein-chemical methods.
Biochemical tests:
SDS-PAGE (purity > 90 %); Western blot with polyclonal rabbit anti-ApoH antibody.
Observed molecular weight (SDS-PAGE):
Approx. 70 kDa
Immunological tests/Functionality: Standard ELISA test (checkerboard analysis of positive/negative sera panels).
Recommended buffer/storage and handling conditions:
Recommendations for storage buffer: ionic strength around 250 mM, neutral to slightly alkaline pH and 20 % glycerol as cryoprotective agent. Storage temperature: –70° to –80° C. Repeated freeze/thaw cycles should be avoided. Use of reducing sulfhydryl reagents and denaturing detergents will result in the destruction of conformational epitopes.
Coating concentration:
0.5-1.0 μg/ml (depending on the type of ELISA plate and coating buffer). Suitable for labeling of functional groups.
Due to the diversity of existing β2-glycoprotein 1 / ApoH coating protocols (e.g. binding to phospholipid-precoated plates) further recommendations cannot be given.
